    Chloroquine and breast cancer

To date, it was assumed that chloroquine increases the sensitivity of cancer cells to chemotherapy by means of a direct effect on the cancer cells. However, a recent study has demonstrated that chloroquine also normalizes the abnormal blood vessels in tumors.

The purpose of this study is to test the hypothesis that chloroquine will reduce the ability of ductal carcinoma in situ DCIS to survive and spread. Participants will receive either chloroquine standard dose 500mg/week or chloroquine low dose 250mg/week for 1 month prior to surgical removal of the tumor. Chloroquine CQ is a well known, well tolerated medication that has been used for many years traditionally for arthritis, lupus, and malaria. It has anti-inflammatory properties but is often the drug of choice for arthritis and malaria due to its few side effects. This medicine may add to the chance of getting some types of cancer. Talk with the doctor. Keep away from children. Accidental exposure may cause death. If a child takes chloroquine by accident, get medical help right away. If you are 65 or older, use chloroquine with care. You could have more side effects.

The anti-cancer effect of the antimalarial agent chloroquine when combined with conventional chemotherapy has been well documented in experimental animal models. This blood vessel normalization results in an increased barrier function on the one hand, and in enhanced tumor perfusion on the other hand, which increases the response of the tumor to chemotherapy.

Chloroquine diphosphate CQ, a worldwide used anti-malarial drug, has recently been focused due to its potential biological effects on cancer cells, such as the inhibition of cell growth and/or induction of cell death in human lung cancer A549 cells, glioma cells, human breast cancer cells, and mouse colon cancer CT26 cells, resulting in anti-cancer effects 3-7.

Chloroquine prevented DCIS xenografts' outgrowth in athymic mice and inhibited N-methyl-N-nitrosurea–induced mammary carcinogenesis, suggesting chloroquine-based therapy as a possible agent in the prevention of initial premalignant lesions from progressing to breast cancer. Jul 15, 2014 Autophagy and cancer. A large body of evidence suggests that the relationship between autophagy and cancer is complex.42, 43 On the one hand, autophagy appears to inhibit malignant transformation, reflecting its capability to limit the accumulation of potentially oncogenic entities like depolarized mitochondria which overproduce potentially genotoxic reactive oxygen species, ROS. For example, chloroquine has demonstrated promising effects as an anti-cancer agent, particularly in breast cancers 16–19. Chloroquine has been shown to inhibit breast cancer growth in vitro, and low doses of chloroquine have induced resistance to mammary carcinogenesis in a rat model of chemically-induced breast cancers 20, 21.
